The medium was developed for the cultivation of normal and neoplastic leukocytes, bone marrow cells and hybridoma. Especially with the addition of fetal bovine serum in various amounts, which provides the cells with growth factors, trace elements and hormones such as insulin, the RPMI medium is suitable for many cell lines.
The medium contains 19 essential and non-essential amino acids, including glycine, 11 vitamins, 6 salts and sodium bicarbonate. It is designed for an environment with 5% carbon dioxide. Its properties make it particularly suitable for cells in suspension or monolayer and it is often used to grow a variety of mammalian cell types.
RPMI Medium 1640 is unique compared to other media because it contains the reducing agent glutathione and high concentrations of vitamins. Which aren’t inclouded in Eagle’s Minimal Essential Medium or Dulbecco’s Modified Eagle Medium.
