BSA zur Stabilisierung von Enzymen

Beschreibung: BSA wird oft mit Restriktionsenzymen geliefert. Es verhindert die Anlagerung des Enzyms an den Wänden der Reaktionsgefäße. BSA stabilisiert auch Proteine während der Inkubation.

Konzentration: 10 mg / ml (1 % ) (oder andere Konzentration auf Anfrage)

Lagerpuffer: 20 mM KPO4, 50 mM NaCl, 0.1 mM EDTA, 5% Glyzerin, pH 7.0 @ 25°C

Qualitätskontrollen:
16-Hour Incubation:
A 50 µl reaction containing 1 µg of FX174 DNA (HaeIII digested) and 50 µg of BSA incubated for 16 hours at 37ºC resulted in a DNA pattern free of detectable nuclease degradation as determined by agarose gel electrophoresis.

RNase Assay:
Incubation of 50 µg of BSA with 1 µg RNA Ladder for 1 hour at 37°C in 50 µl assay buffer [70 mM Tris-HCI, 10 mM MgCl2, 5 mM DTT, (pH 7.6, 25°C)] followed by agarose gel electrophoresis resulted in no detectable RNA degradation.

Endonuclease Assay:
Incubation of 50 µg of BSA with 1 µg FX174 RF I DNA for 4 hours at 37°C in 50 µl of 1X Reactionbuffer 2 resulted in < 10% conversion to RF II.
